Unable to Assign Resources to a Settlement
Players cannot assign resources to a Settlement that doesn't have an available Trade Route. However, a tooltip message doesn't appear to warn the player of this.

This should now be fixed with Update 1.1.1.
This is currently planned to be addressed in a future update.
